چکیده

The ventricular zone (VZ) of the embryonic dorsal telencephalon is a major site for generating cortical projection neurons. The transcription factor Pax6 is expressed in apical progenitors (APs) residing in the VZ in a rostrolateral-high to caudomediallow gradient and its levels are highest at early stages of corticogenesis. Previous studies mainly focused on Pax6 / mice have implicated Pax6 in cortical progenitor proliferation, migration, neuronal differentiation and formation of superficial cortical layers. We wish to explore the developmental significance of Pax6 expression levels in cortical progenitors at different developmental stages and levels across the rostral-caudal axis of the cortex. In order to address this issue, we are using a floxed Pax6 allele, where the exposure to Cre recombinase results in excision of Pax6 and initiation of YFP expression. Use of a tamoxifen-inducible Emx1-CreER driver allows us to remove the gene from cortical progenitors in a mosaic manner at times coincident with deepor superficial layer neurons being generated. Our preliminary results suggest that when Pax6 is removed from cortical progenitors at mid-stages of corticogenesis, the thickness of the postnatal cortex is severely reduced due to a remarkable under-production of superficial layer neurons. Deletion of Pax6 at late stages of corticogenesis results in the contribution of late-born Pax6 null cells to deeper laminar positions compared to Pax6 cells. This suggests that the normal low expression levels of Pax6 in late APs might have an effect on the specification of superficial layer neurons.

ML1419c peptide immunization induces Mycobacterium leprae-specific HLA-A*0201-restricted CTL in vivo with potential to kill live mycobacteria.

MHC class I-restricted CD8(+) T cells play an important role in protective immunity against mycobacteria. Previously, we showed that p113-121, derived from Mycobacterium leprae protein ML1419c, induced significant IFN-γ production by CD8(+) T cells in 90% of paucibacillary leprosy patients and in 80% of multibacillary patients' contacts, demonstrating induction of M. leprae-specific CD8(+) T ce...

Blockade and stimulation of renal, adrenal, and vascular angiotensin II receptors with 1-Sar, 8-Ala angiotensin II in normal man.

We have assessed the capacity of an analogue of angiotensin II (A II), 1-Sar, 8-Ala A II (P113) in normal man to stimulate and block responses to A II in four systems: blood pressure was monitored directly from an arterial catheter, and renal blood flow was measured with 133Xe and arterial renin and aldosterone concentrations by radioimmunoassay. The 31 normal subjects were in balance on a dail...

P113 is a merozoite surface protein that binds the N terminus of Plasmodium falciparum RH5

Invasion of erythrocytes by Plasmodium falciparum merozoites is necessary for malaria pathogenesis and is therefore a primary target for vaccine development. RH5 is a leading subunit vaccine candidate because anti-RH5 antibodies inhibit parasite growth and the interaction with its erythrocyte receptor basigin is essential for invasion. RH5 is secreted, complexes with other parasite proteins inc...

Angiotensin antagonists with increased specificity for the renal vasculature.

This study was designed to ascertain whether renal vascular angiotensin receptors differ from other systemic angiotensin receptors and whether, on that basis, antagonists with greater specificity for the renal vasculature can be defined. Femoral and renal blood flow and their responses to angiotensin II (AII) and its heptapeptide analogue, 1-des Asp AII (AIII), were measured with an electromagn...

Ontogeny of poly(ADP-ribose) polymerase-1 in lung and developmental implications.

Poly(ADP-ribose) polymerase 1 (PARP-1) is the predominant NAD-dependent modifying enzyme in DNA repair, transcription, and apoptosis; its involvement in development has not been defined. Here, we report expression and cellular localization of PARP-1 in developing rat and human fetal lung, in vivo and in explant culture, and effects of inhibiting PARP-1 activity on lung surfactant protein (SP) e...
